Microbial growth in industrial organic chemicals can present a major problem. If left unchecked, microbial contamination in raw materials and intermediate products can compromise manufacturing process performance and equipment integrity. These same sources can also impact final product quality. The best solution is early, accurate detection for proactive rather than reactive treatment. The QGO-M XLPD test kit utilizes a 3-minute filtration-based analysis to measure a parameter called cellular ATP (cATP™).
cATP represents ATP from living microorganisms in suspension in a fluid and therefore is a direct indication of the planktonic population.
Intracellular ATP (cATP), that is the ATP contained in living biological cells, is the true indicator of the concentration of living microorganisms in the sample. Measuring living biomass through the cATP parameter includes measuring culturable, non-culturable and difficult to culture bacteria.
Increase of intracellular ATP in samples indicates proliferation of microorganisms and a quality degradation of complex organic samples. If active biomass reaches levels above recommended thresholds, a microbiological drift is taking place. Increase of the active biomass can be seen as an early alarm of microbial disorders in the installation.
Organic compounds are washed from the sample for their elimination. The sample is submitted to filtration through a filter attached to a syringe (QGO Prep kit). Only living micro-organisms are retained by the filter’s porosity. Then, lysis of microorganisms is performed directly on the filter using the UltraLyse™ reagent and intracellular ATP is released. The ATP solution is then diluted and the concentration of ATP is measured using Luminase™ reagent. An external standard calibrated ATP solution, Ultracheck™ 1, allows providing reliable quantitative results.
Total time to result is 3 minutes. Final results are delivered either in pg ATP/ml or Equivalent Microorganisms/ mL (1pg ATP = 1000 Equivalent microorganisms). Equivalent microorganisms include culturable microorganisms and non culturable microorganisms and this level is not directly comparable to CFU results from culture methods.
QGO-M XLPD is available for samples that are more difficult to filter and/or have large quantities of inhibitory components (such as latex, polymers, concrete admixtures and personal care products). Therefore, QGO-M XLPD contains same components as QGO-M but QGO-M XLPD includes also 1/10 initial pre-dilution of the sample and standard Luminase™ is replaced by LuminaseXL (10 times stronger than standard Luminase).
